چکیده

The lactating rabbit synthesizes milk triacylglycerol, which contains a high proportion (approx. 65 mo1/100mol) of medium-chain fatty acids, the remainder being long-chain fatty acids (Smith et al., 1968). Experiments in vivo with suckling rabbits had shown that medium-chain milk fatty acids were transported to the liver via the hepatic portal vein, and were taken up by the liver. In order to compare the metabolism of mediumand long-chain fatty acids by the liver, octanoate (C8:o) and palmitate (C16:o) were incubated at a concentration of 1 mM with hepatocytes obtained from 10-1 2-day-old rabbits and rates of oxidation to CO, and of ketone-body production were measured. This is a period of development when ketone bodies could be used for lipid synthesis in the brain, as has been shown for the suckling rat (Webber & Edmond, 1979). Both fatty acids were found to be ketogenic in hepatocytes, though octanoate produced about 60% more acetoacetate plus ~-(-)-3-hydroxybutyrate than did palmitate (Fig. 1). This synthesis of ketone bodies represents a major route of

Metabolic fate of non-esterified fatty acids in isolated hepatocytes from newborn and young pigs. Evidence for a limited capacity for oxidation and increased capacity for esterification.

In hepatocytes isolated from 48 h-old starved of suckling newborn pigs or from 15-day-old starved piglets, the rate of ketogenesis from oleate or from octanoate is very low. This is not due to an inappropriate fatty acid uptake by the isolated liver cells, but results from a limited capacity for fatty acid oxidation. Some 80-95% of oleate taken up is converted into esterified fats, whatever the...

Suckling-Weanling Transition in the Rat STUDIES WITH ISOLATED HEPATOCYTES By MANUEL BENITO, EMMA WHITELAW* and DERMOT

The rates of ketogenesis from endogenous substrates, butyrate or oleate, have been measured in isolated hepatocytes from suckling and weanling rats. Ketogenesis from endogenous substrate and from oleate decreased on weaning, whereas the rate from butyrate remained unchanged. It is concluded that the major site of regulation of ketogenesis during this period of development involves the disposal ...

Relationship between ketogenesis and gluconeogenesis in isolated hepatocytes from newborn rats.

In hepatocytes from 1-day-old rats, active gluconeogenesis occurs in parallel with active ketogenesis, although the carbon atoms of non-esterified fatty acids do not participate in glucose synthesis. Once a significant ketogenesis is established, a further increase does not enhance gluconeogenesis. Indeed, octanoate is more ketogenic than oleate, but stimulates gluconeogenesis to a similar extent.

Interrelations between C4 ketogenesis, C5 ketogenesis, and anaplerosis in the perfused rat liver.

We investigated the interrelations between C(4) ketogenesis (production of beta-hydroxybutyrate + acetoacetate), C(5) ketogenesis (production of beta-hydroxypentanoate + beta-ketopentanoate), and anaplerosis in isolated rat livers perfused with (13)C-labeled octanoate, heptanoate, or propionate. Mass isotopomer analysis of C(4) and C(5) ketone bodies and of related acyl-CoA esters reveal that C...

Development of fatty acid oxidation in neonatal guinea-pig liver.

The capacity of foetal and neonatal liver to oxidize short-, medium- and long-chain fatty acids was studied in the guinea pig. Liver mitochondria from foetal and newborn animals were unable to synthesize ketone bodies from octanoate, but octanoylcarnitine and palmitoylcarnitine were readily ketogenic. The ketogenic capacity at 24 h after birth was as high as in adult animals. Hepatocytes isolat...
